コロンブスが幸福であったのは、彼がアメリカを発見した時ではなく、 それを発見しつつあった時である。 幸福とは生活の絶え間なき永遠の探求にあるのであって、断じて発見にあるのではない。

Columbus was not happy when he had discovered Americas, but rather when he was just about to discover it.
Happiness [exists in/lies on] the contineous endeavour, certainly not [in/on] the discovery.
[exists in]を使うなら、「～の中に存在する」となり、その行為そのものにという意味合いになると思います。
それに対して、[lies on]を使うなら、「～の上に横たわる」となり、その行為が幸せを運ぶ未来へのレールだというようなニュアンスになるかと。

Columbus would have find a happiness when he was discovering America, not when he discovered it. Happiness lies in the searching activity itself, not in the discovery.
注）
コロンブスがアメリカ大陸を「発見」する以前に、アメリカ大陸にはアジアから移動してきた先住民族が住んでいました。また、コロンブスよりも先に、北欧からレイフ・エリクソンらがアメリカ大陸に到達しています。従って、「コロンブスがアメリカ大陸を発見した」と表現するよりも、「大西洋航路を発見した」・「アメリカ大陸を再発見した（rediscover）」・「西欧からアメリカ大陸に初めて到達した（reach）」などと表現するほうが正確なようです。
